What is the difference between for example a £300 road bike and a £500 road bike?

The more expensive bikes have lighter frames and better components. A £300 bike is likely to have an aluminum frame with a carbon fork, low end Mavic wheels, and Shimano Sora or Tiagra components. You could find a £500 bike with a carbon fiber frame, Mavic Aksium wheels, and Shimano 105 components, for example. The benefit of a lighter frame and wheels is really noticeable when you are going up a hill. The more expensive components are lighter, work smoother, and hold their adjustments longer.

Answer by curious georgeWell, $ 500 still isn’t a ton of money for a road bike. The differences between a cheap and expensive road bike are little subtle things. An expensive bike might be made of carbon fiber which is much lighter and absorbs shocks more, whereas a cheap bike might be made of steel or aluminum. Expensive bikes might have components like Shimano Ultegra or Dura Ace, whereas the cheap one might have Shimano Sora parts. The higher end components shift better, are better quality, and usually last longer. Also, some people don’t really know the difference but just buy the expensive bikes for the name thing. If a bike is used, that doesn’t necessarily make it worse than a brand new bike. It all depends on the previous owner. Some owners never lube their bike and trash it out, so obviously it might be better to get a new bike. On the other hand, some owners not only lube it, but take it apart to clean it, maybe even upgrade some parts.
